The actress, 43, revealed her growing baby bump to the world as she walked the red carpet at the Odeon Luxe in Leicester Square.
She joined her husband, film producer Jasper Waller-Bridge, 37, as they announced their happy news.
Michelle arrived in a powder blue off-the-shoulder ballgown with a fitted bodice as she showed off her bump.
She has portrayed fan-favourite Lady Mary Crawley since the period drama debuted on ITV in 2010.
Arriving for the afterparty, Michelle changed outfit and sported a comfortable long black dress that she paired with a white long coat.

Meanwhile, Nadiya Bychkova beamed as she was pictured leaving the UK premiere of Downton Abbey: The Grand Finale on Wednesday night following her fall.
The Strictly Coming Dancing professional, 36, was snapped smiling as she left the Odeon Luxe Leicester Square.
She appeared to be in good spirits following the movie despite the miserable rainy whether for the red carpet, as she dazzled in her white evening gown.
The striking white frock exuded elegance and sophistication and featured a plunging V-neckline and long sleeves.
Nadiya’s Strictly co-star Neil Jones was also spotted leaving the premiere on Wednesday evening, looking dapper in a navy suit, though showed he was the perfect gentleman by giving his jacket to partner Chyna Mills.
Also snapped leaving were Verity Bowditch who wowed in a brown asymmetric gown and Emma Walsh who donned a black off-the-shoulder gown.

It was previously revealed how Strictly star Nadiya was taken to hospital on Tuesday after falling and landing on her back during rehearsals.
She was training in north London when she was left in agony after injuring her back, with crew forced to phone an ambulance.
Nadiya later shared a picture from her hospital as she updated fans on her injury – and revealed if she would be able to dance again soon.
She wrote: ‘Well… where do I start? What a 24 hours! Thank you so much for all the messages, love and support. I just wanted you to know that I am ok.

Nadiya continued: ‘I’m smiling in the picture but it was really scary and there were a few tears as I had to lie still for hours but I was given the all-clear after an x-ray and allowed to go home.
‘I’ve got some physio today but I will be back on that dance floor soon and I’ll be ready to shine and sparkle in the new series of the biggest show on TV.
‘I had a fall in training yesterday and landed on my back. I was really worried for a while and I was so thankful to the Strictly family and medical team who really looked after me.
‘The staff at the hospital were so kind and caring.’
